Neurobiopieltec-CM Project Phases with TERMeG Group Participation (Carlos III University of Madrid)

Phase 1:

* Studies on the induction of psoriatic phenotype in 2D.
* Development of in vitro skin models.
* Development of an organ-on-chip system for both skin models.

Phase 2:

* In vitro model of psoriatic skin in a static system.

Phase 3:

* Integration of skin models (healthy and psoriatic) into dynamic organ-on-chip systems.

Phase 4:

* Study of therapeutic molecules in in vitro skin models integrated into dynamic systems for psoriasis treatment.

Activities within this phase:

The TERMeG research group participates in objectives 1 and 3 of Phase 1 of the project.

On one hand, they are involved in the design, development, and manufacturing of a dynamic and microphysiological organ-on-chip (OoC) system that allows for the generation of an in vitro skin model. Different design alternatives for integrating the in vitro psoriatic skin model into the dynamic culture system will be studied.

In parallel, studies on the induction of the psoriatic phenotype in epidermal cells (human keratinocytes) will begin, using both immortalized cell lines and primary cultures. For this purpose, various pro-inflammatory cytokines will be studied at different concentrations and time points. Subsequently, phenotype expression studies will be conducted by immunocytochemistry, and gene expression studies by qPCR.

Tasks to be performed:

* Cell culture, including both immortalized cell lines and primary cells.
* Development of 3D cultures of in vitro human skin tissue.
* Development of an in vitro psoriatic skin model and evaluation of inflammation markers.
* Gene expression analysis in healthy and psoriatic skin models through qPCR, Western Blot, ELISA kits, immunofluorescence, and immunohistochemistry.
* Design and manufacturing of a microphysiological system for the development of a skin-on-chip model that can house biosensors.
* Automation of processes, including programming, sensors, bioreactors, and microfluidic systems.

This contract is funded by the Community of Madrid through the project "Integration of New Organ-on-chip Technologies for the Study of Skin and Neurodegenerative Diseases," with the acronym NeuroBiopieltec-CM, and reference TEC-2024 BIO-98. It was awarded under the 2024 R&D Activities Programs in Technologies call by the Directorate General for Research and Technological Innovation, via ORDER 5696/2024, of December 10, from the Minister of Education, Science and Universities.

* Degree in Bioengineering or Engineering with a strong multidisciplinary profile, demonstrating knowledge of cell and molecular biology, biomaterials, or biophysics.
* A Master's degree in these fields and professional and/or research experience in these areas will be favorably valued.
* Essential to have demonstrable experience in tissue engineering, specifically in skin.
* Essential to have knowledge of microphysiological systems or organ-on-chip models, more specifically in skin.
* Experience in electronics and instrument programming for process automation and optimization is a plus.
